abstract = {{The digitization of the industry, the drive towards smart factories as well as the Internet of Production (IoP) require rising smartness of products and services. Smart physical products are often mechatronic products that include increasing amounts of software. The development of software, however, comes along with new challenges for companies specialized in developing mechanical, electrical or electronic products. Some of these challenges address the product lifecycle management (PLM)-related business and work processes. The management of software lifecycles requires a much more rigorous requirements management. Furthermore, special solutions for management of source code in distributed development teams are needed. The build-process and testing activities need to be conducted in a systematic manner. The generation and provision of different licensing models need to be mastered and finally the issue of security needs to be addressed for any product that can be networked---which by the way is a strategic target of nearly any product developing company. Application Lifecycle Management (ALM) covers many of the above-mentioned issues. IT solutions for ALM are comparable to traditional PLM solutions, but focus particularly on software as a product. Thus, these systems have become widely used by software companies in the same manner as PLM solutions belong to the standard enterprise IT environment of companies developing physical products. With software penetrating traditional physical products, product managers, product developers, manufacturing staff etc. need to work with both, PLM and ALM, since neither solution is able to cover both domains sufficiently. However, ALM and PLM solutions feature redundant functionality. Thus, best practices for the systematic integration of ALM and PLM are required.}},

abstract = {{The physical response of lakes to climate warming is regionally variable and highly dependent on individual lake characteristics, making generalizations about their development difficult. To qualify the role of individual lake characteristics in their response to regionally homogeneous warming, we simulated temperature, ice cover, and mixing in four intensively studied German lakes of varying morphology and mixing regime with a one-dimensional lake model. We forced the model with an ensemble of 12 climate projections (RCP4.5) up to 2100. The lakes were projected to warm at 0.10–0.11 ∘C decade−1, which is 75 %–90 % of the projected air temperature trend. In simulations, surface temperatures increased strongly in winter and spring, but little or not at all in summer and autumn. Mean bottom temperatures were projected to increase in all lakes, with steeper trends in winter and in shallower lakes. Modelled ice thaw and summer stratification advanced by 1.5–2.2 and 1.4–1.8 days decade−1 respectively, whereas autumn turnover and winter freeze timing was less sensitive. The projected summer mixed-layer depth was unaffected by warming but sensitive to changes in water transparency. By mid-century, the frequency of ice and stratification-free winters was projected to increase by about 20 %, making ice cover rare and shifting the two deeper dimictic lakes to a predominantly monomictic regime. The polymictic lake was unlikely to become dimictic by the end of the century. A sensitivity analysis predicted that decreasing transparency would dampen the effect of warming on mean temperature but amplify its effect on stratification. However, this interaction was only predicted to occur in clear lakes, and not in the study lakes at their historical transparency. Not only lake morphology, but also mixing regime determines how heat is stored and ultimately how lakes respond to climate warming. Seasonal differences in climate warming rates are thus important and require more attention.}},

N2 can be found as a component besides CO2 in possible feed gases like mine gas, weak gas, or steel mill gas. To determine whether there is an effect on the biological methanation of CO2 and H2 from renewable sources or not, the process was investigated using feed gases containing CO2, H2, and N2 in different ratios, depending on the CO2 content. A possible effect can be a lowered conversion rate of CO2 and H2 to CH4. Feed gases containing up to 47N2 were investigated. The conversion of hydrogen and carbon dioxide was possible with a conversion rate of up to 91 but was limited by the amount of H2 when feeding a stoichiometric ratio of 4:1 and not by adding N2 to the feed gas.
}},

Kinetic tests revealed that the organics in spent grains PW had much lower molecular weights than organics in fine mulch PW. Moreover, isotherms showed that they were more strongly adsorbable. This was confirmed in GAC column experiments, where the breakthrough curves could be predicted fairly well by a dynamic adsorption model. On the other hand, ozonation had a stronger effect on fine mulch PW with respect to an enhancement of the aerobic degradability. Thus, the type of input material determines the properties of soluble reaction products from the carbonization process that must be accounted for when selecting the most suitable post-treatment method for HTC PW. However, adsorption on granular activated carbon should always be the final stage.}},

abstract = {{The concepts for the minimum dwelling investigated by inter-war modernists were further developed and largely applied in the construction of postwar large-scale housing. As elsewhere in post-war Europe, affordable housing was
high on the agenda in Socialist Yugoslavia. The right to a residence was an imperative of the socialist state, which set an enormous housing construction program so that each family could be housed in its own apartment. To meet the huge
housing needs, another imperative was to build quickly and cheaply. New Belgrade, a project for the capital of the newly founded socialist state, eventually became the biggest construction field for providing societally owned flats for tens of thousands of inhabitants. The demand for huge amounts of flats, efficient construction and lowcosts dictated the optimization of design, standardization, and rationalization. The paper investigates the design of New Belgrade flats focusing on different aspects of the “minimum” that were applied. It additionally analyses how compared to the interwar
concepts the perspective on the minimal needs changed. Furthermore, it compares these standards and needs with the actual ones. The research aims to trace these changing perspectives on minimum, to rethink the modernist minimum
dwelling and explore how it relates and reflects the minimum in design today. }},

A considerable part of machine learning research focuses on the selection of relevant, non-redundant features. This contribution details an approach to group and fuse redundant features prior to learning and classification. Features are grouped relying on a correlation-based redundancy measure. The fusion of features is guided by determining the majority observation based on possibility distributions. Furthermore, this paper studies the effects of feature fusion on the robustness and performance of classification with a focus on industrial applications. The approach is statistically evaluated on public datasets in comparison to classification on selected features only.}},

Monitoring the curing process of polymers has the potential to decrease repair costs by time optimisation and quality control. In this paper Lamb waves are utilised to determine the degree of cure. Waves are excited and recorded by two piezoelectric transducers, one serving as an actuator and the other as a sensor. The recorded signals are processed with a complex wavelet transform, which allows more accurate feature extraction than calculating features in time domain. Extracted features are the transmitted signal energy of the wave and the resonance frequency of the curing polymer. Waves are excited at different frequencies to identify the current resonance frequency. Excitation at or near the current resonant frequency ensures that curing is monitored with maximum sensitivity.}},

Cash supply is very important for the worldwide economy. Within the supply chain, handling processes are highly automated, e.g., automated teller machines are installed to ensure an efficient and reliable cash supply. Recently, there is a trend in automated cash handling systems to replace the cash cassettes for each denomination by a simple pitch-in-plastic-bag for all banknotes. This necessitates new automated handling approaches because the bag content will unload irregularly. We propose a new approach which is able to detect the pose and possible occlusions of randomly distributed textured banknotes with simple smartphone cameras. This information can be used to control a handling robot-picking out banknotes.}},

abstract = {{To connect terminals in a cyber–physical system, large quantities of electrical contacts are used. In order to guarantee a high reliability of the system, the lifetime of the electrical contacts should be very long. Thus, it is of great importance to understand the failure mechanism and then to predict the lifetime of the electrical contacts. For the applications under high thermal and/or mechanical loads, noble plating is a good choice, considering its inertness to oxidation. For noble plating, one of the most critical failure mechanisms is the fretting wear. Wear debris generated in the contact area, acting as the third bodies, will greatly influence the further wear behavior and electrical performance. In this study, the state of the art regarding third bodies is firstly reviewed, and then the influence of the third bodies on the wear and electrical performance is investigated, from the aspects of lifetime and the element distributions in contact area. Finally, an example of prediction of the wear of noble plating is shown with the consideration of the third bodies. Based on this study, by involving the third bodies, the wear of noble plating can be predicted with a higher accuracy.}},

abstract = {{The control and structural expansion of decentralized energy systems are very challenging due to the volatility of renewable energies and progressive structural changes. For balancing out seasonal fluctuations, conversions into heat or gas in combination with long-term storages are frequently discussed approaches. In context of an optimal conceptual synthesis of such systems, investigations regarding the operation and design require a large time period of at least one year. In order to solve such optimal control problems, an immense calculation time is required. This contribution presents a multistep approach which determines the optimal operation strategy in an iterative way and is capable of reducing the calculation effort. In the first step, a rough optimization incorporating a low modelling depth is performed. Especially in combination with a rough time discretization, dynamic short-term storages (e.g. electrical batteries) can become irrelevant from an optimization point of view. Therefore, the considered system can be virtually reduced by several state and control variables resulting in a significantly reduced computation time. In a second optimization, the optimal control problem is constrained using the results of the previous step. Especially the obtained values for the state of charge of the long-term storage improve significantly the quality of the second optimization. While in the first step, the dynamic programming is utilized to solve the optimal control problem in one instance, the second step uses the mixed integer linear programming to solve multiple short time periods of the optimal control problem in a sequential way. Results are presented on the basis of a simple test scenario where the electrical energy supply of a residential quarter is investigated using real photovoltaic data of one year, a modelled fuel cell system as long-term storage and an electrical battery storage as short-term storage.}},

Hence, work assistance has emerged as an important theme in the context of cyber-physical production systems and Industry 4.0 to assist workers in assembly, logistics, maintenance and supervision. Recent research in this domain has focused on demonstrating assistance applications using mobile computing devices such as tablets, smartphones, AR/VR glasses and wearables, but the aspects of technology induced complexity in industrial work distribution, concurrency, information complexity, and variability of information interaction, and their subsequent effect on human workers is yet to be tackled. This paper has two core contributions: first, it reframes the problem of complex industrial work through activity theory, which leads to a conceptual model that couples human information needs to interactive artefacts within an activity context. Second, the problem of assistance is viewed as managing information flow between multiple devices grouped into fluid and adaptive activity contexts, managed by MiWSICx, (Middleware for Work Support in Industrial Contexts) a novel, distributed middleware designed using the actor model of concurrent computation.}},

Such systems utilize augmented- and virtual-reality for operator training and live guidance as well as mobile maintenance and support. This is particularly important in the modern production reality of ever-changing products and `lot size one' customization of production. This paper focuses on the application of machine learning approach to extend the functionality of assistance systems. Machine learning provides tools to analyse large amounts of data and extract meaningful information. The goal here is to recognize the movement of an operator which would enable automatic display of instructions relevant to them.We present the challenges facing machine learning applications in human-centered assistance systems and a framework to assess machine learning approaches feasible for this scenario. The approach is assessed on a historical data set and then deployed in a work station for live testing. The post-hoc, or historical, analysis yields promising results. The ad-hoc, or live, analysis is a complex task and the results are affected by multiple factors, most of which are introduced by the human influence. The contribution of this paper is an approach to adapt state- of-the-art machine learning to operator movement recognition with a special focus on approaches to spatial time series data pre-processing. Presented experiment results validate the approach and show that it performs well in a real-world scenario.}},

However, at this point it is still unclear which problems of information representation actually arise in manual assembly operations and how these problems may be solved through the use of informational assistance systems. In two example cases, this paper identifies problems of information representation by juxtaposing actual and target processes. Using the method of inductive category development, the identified problems of information representation are collected in a total of five categories, for which principles for the design of informational assembly assistance systems are then formulated.}},

Assembly assistance systems supply employees with information according to their needs and individual situation. This article aims to present important principles for the design of informational assembly assistance systems. The empirical basis for these principles is formed by projects involving the introduction of informational assistance systems for assembly work. The trends and design recommendations are explained using a model, which illustrates important associations between the complexity of assembly tasks, the demands on the mental capacity of employees, work productivity and the use of assembly assistance systems.}},

abstract = {{The German regulation for the protection of surface waters (OGewV) supplements and substantiate the requirements regarding the implementation of the Water Framework Directive (WFD) and contains detailed regulations regarding the classification, presentation and monitoring of the ecological and the chemical status of surface waters including thresholds for phosphorus. The quality of Germany’s water bodies has greatly improved in recent decades. Despite the considerable advances that have been made in the sewage treatment domain, point source pollution from certain nutrients and pollutants remains unduly high and thus needs to be reduced. This applies, for example, to phosphorus, as well as micro-pollutants (MPs) that in many cases cannot be adequately eliminated at wastewater treatment plants. In addition such inputs from combined sewer and rainwater (separate sewer) overflow (CSO/SSO) are also still a problem in certain water bodies.
Sub-surface vertical flow constructed wetlands (SSVF) have been used since the late 1990s for CSO/SSO because of its high mechanical and biological efficiency. To achieve in particular a stable and long-term phosphorus reduction iron-containing substances are blended so far to large-scale SSVF as additives during the construction. However, some disadvantages in this type of substrate improvement must be stated. High and thereby costly material and installation costs are required and a subsequent melioration of a SSVF under continuous operation is impossible. The only possibility exists in the cost-intensive replacement of the entire soil body including replanting. All disadvantages can be avoided if instead of a substrate melioration a separate sorption stage, which is filled exclusively with iron-containing additives, is connected downstream of the SSVF. The material consumption is limited only to this sorption stage. This paper gives the results of an appropriate 3 year large-scale investigation including reduction rates and operational data concerning the standard parameters with the main focus on phosphorus as well as selected MPs and selected heavy metals as four of the six heavy metals investigated are classified as priority or even priority hazardous substances.
The sorption stage consisted of two reactors, operated in series. The first reactor was filled with FerroSorp® (granular iron hydroxide), the second reactor with GAC. The P-removal of SSVF and sorption stage is in a load-related magnitude of 94% (oPO4-P), resulting in concentrations mainly less than 0.1 mg oPO4-P/L. Considering the FerroSorp® load capacity of 40% after two years of operation it can be estimated that a separated sorption stage might achieve a minimum lifetime of 4 years. Current German standards for meliorated SSVF generally recommend an amount of filter substrate of 10% by mass to achieve P-removal rates of 80% for two years of lifetime. This lifetime is half as long as with the investigated downstream sorption process and this with the thirtyfold increased use of iron-containing additive. Commenting the MP elimination, a substantial MP-loading of the CSO as well as a substantial MP-removal in the SSVF can be stated. Ten out of eleven MPs are reduced by the SSVF between 20% up to 80% (Metoprolol). The final downstream GAC sorption reactor eliminated all MPs almost completely as expected. The analyzed heavy metals cadmium (Cd), copper (Cu), nickel (Ni), lead (Pb) and zinc (Zn) are retained in the entire three-stage plant (SSVF, two sorption reactors) with high efficiencies between 80% and 97%. Mercury (Hg) was analyzed only at very low concentration level. }},

abstract = {{This paper presents the empirical validation of the time accuracy of the process building block system MTM-HWD® (Human Work Design). The analysis is based on process data collected from 62 different workplaces in German manufacturing companies. The accuracy of the process system is analyzed by studying the frequency of employed process building blocks in the data collection procedure. Subsequently, the differences between the MTM-HWD® and MTM-1 cycle times are evaluated. The findings show that there is no significant difference from samples of prior MTM-1 studies with respect to the frequency of employed MTM-1 process building blocks. It is shown that there is a systematic difference and a linear relationship between the MTM-HWD® and MTM-1 time data. However, it is demonstrated that the relatively larger MTM-HWD® times do not differ more than 5 % from the MTM-1 times when considering the 95 % confidence interval of the mean. These results meet the development aims in terms of the defined accuracy of the method of ± 5 % in comparison to MTM-1.}},

The International Conference in Berlin takes the 100th anniversary of the Bauhaus as an opportunity to discuss the significance of modernity in the 21st century. The conference focus will be on the concepts, visions, and impulses emanating from Modern Movement and how they can be related to today’s social, economic, cultural and in particular creative issues.

Are the social, spatial and constructional concepts formulated by modern movement and post-war modernism still sustainable today?
What role do cultural and climatic conditions play in the preservation, renovation and transformation of spaces, buildings, and modern
movement sites?
How can the basic ideas of classical modernism be continued 100 years later and thus contribute to solving current challenges?
What contribution can be expected from academic and professional education, and which learning formats are suitable for this?

The 2019 DOCOMOMO Germany event will move from Karlsruhe and be held for the first time in Berlin, Neukölln at the Werkstatt der Kulturen. It continues the tradition of the Karlsruhe DOCOMOMO Germany Conference. This year the conference is co-organised by ‘RMB‘, a project that is funded by the EU and coordinated by the OWL University of Applied Sciences. RMB initiates a pedagogical framework on a European level on the reuse of modernist buildings based on common definitions, methods, and approaches. RMB prepares a Joint Master on Reuse of Modernist Buildings. This cooperation of DOCOMOMO Germany and RMB resulted in a new conference format: a combination of invited keynote speakers and selected scientific lectures under the theme of ‘What interest do we take in the Modern Movement today?‘.}},

Improper or lack of maintenance may result in excessive component wear, production quality deterioration, or even longer downtime. However, today's production facilities strive to devote the least amount of necessary maintenance time in order to maximize production time. Therefore, new solutions for deliberate and efficient maintenance are needed. The solution proposed in this paper benefits from the newest trends and innovations in industry, namely the Asset Administration Shell (AAS) which is part of the Industrie 4.0 (I4.0) concept. The AAS shall contain the maintenance submodel which shall be used for supporting humans during the maintenance process. The submodel provides a standardized description of required tools and parts as well as step-by-step instructions which also include safety concerns and multimedia files, such as pictures and videos. In this way, maintenance can be carried out more reliably, resulting in reduced downtime. In addition, feedback from the maintenance process shall be stored in the submodel and fed through an I4.0-compliant network to other processes from different phases of the life cycle in order to improve them.}},

This enables the usage of data, to create additional benefit, such as condition monitoring or optimization. These capabilities can lead to cognition, such that the system is able to adapt independently to changing circumstances by learning from additional sensors information. Developing a reference architecture for the design of CPPS and standardization of machines and software interfaces is crucial to enable compatibility of data usage between different machine models and vendors. This paper analysis existing reference architecture regarding their cognitive abilities, based on requirements that are derived from three different use cases. The results from the evaluation of the reference architectures, which include two instances that stem from the field of cognitive science, reveal a gap in the applicability of the architectures regarding the generalizability and the level of abstraction. While reference architectures from the field of automation are suitable to address use case specific requirements, and do not address the general requirements, especially w.r.t. adaptability, the examples from the field of cognitive science are well usable to reach a high level of adaption and cognition. It is desirable to merge advantages of both classes of architectures to address challenges in the field of CPPS in Industrie 4.0.}},

abstract = {{The glazed envelopes on buildings play a major role in operational energy consumption as they define the boundary conditions between climate and thermal comfort. Such a facade is viewed as an uncontrolled load that sets the operational performance requirements for artificial lighting and air-cooling mechanical systems. This is in contrast to nature, which has evolved materials with the ability to learn and adapt to a micro-environment through selfregulation using materials that are multifunctional, formed by chemical composition in response to solar load. Leaf vasculature formations are of particular interest to this paper. Through leaf maximisation of daylight capture, the total leaf area density and angular distribution of leaf surfaces define the tree structure. This paper will define an approach to simulate nature to advance a microfluidic platform as a dynamic NIR absorber for solar modulation:a transformable network of multi-microchannel geometry matrix structures for autonomous transparent surfaces, for real timeflow management of conductivity. This is realised through active volumetric flows within a capillary network of circulation fluidicswithin it, through it, and out of it for energy capture and storage, the cycle of which is determined through precise management of heatflow transport within a material. This advances transparent facades into an energy system for heat load modulation nested to climateand solar exposure, which is demonstrated in this paper.}},

@article{5435,
abstract = {{Towards renewable energy systems, the coupling of multiple sectors is important and incorporates novel technologies where currently no models exist that correctly represent all transient effects. Therefore, we present a method that incorporates Hardware-in-the-Loop simulations where virtual components as models are coupled to real and experimental facilities in real time. By including experimental components, a higher validity can be obtained and the practical applicability of renewable energy scenario can be discussed more profoundly. In this paper, the considered energy system consists of an experimental biocatalytic methanation reactor, a real photovoltaic park, a regenerative fuel cell and short-term storage units to supply a residential district. A representative control sequence of the methanator is obtained by modeling the scenario as an optimal control problem. A first HIL simulation highlights that modifications of the instrumentation are required for a grid injection of the generated methane. The scientific approach can be applied to any energy system where some of the considered components are available as experimental or real facilities. Non-exisiting components are simply replaced by models. The presented approach helps to determine which parts or process parameters are crucial for the planed operation before the overall energy system is realized on a larger scale. (C) 2019 Elsevier Ltd. Proficiency in the fabrication of highly complex parts forced this astonishing development. This research is based on the idea that through the integration of additive and conventional manufacturing technologies it is possible to achieve great cost and operational benefits especially in the field of tool making for injection molding. Such an integrated manufacturing solution could overcome the limitations of independent additive, subtractive, and post-processing procedures by strengthening their potentialities. The present study highlights the opportunities of a synergy between the above-mentioned manufacturing technologies for the optimized fabrication of injection molds. An additive manufacturing process chain is presented, and special attention is given to the surface quality and its optimization directly in the Selective Laser Melting process. The potentialities of the Laser Surface Re-melting technique are analyzed, and the process optimization leads to a reduction of 45% of the average roughness directly in the SLM process. abstract = {{The increasing amount of alarms and information for an operator in a modern plant becomes a significant safety risk. Although the notifications are a valuable support, they also lead to the curse of overloading with information for the operator. Due to the huge amount of alarms it is almost impossible to separate the crucial information from the insignificant ones. Therefore, new procedures are required to reduce these alarm floods and support the operator to minimize the safety risk. One approach is based on learning a causal model that represents the relationships between the alarms. This allows alarm sequences that are causally implied to be reduced to the root cause alarm. Fundamental element of this approach is the causal model. Therefore in this work, different probabilistic graphical models are considered and evaluated on the basis of appropriate criteria. A real use case of a bottle filling module serves as a benchmark for how well they are suitable as a causal model for the application in alarm flood reduction.}},

abstract = {{Along with the constantly increasing complexity of industrial automation systems, machine learning methods have been widely applied to detecting abnormal states in such systems. Anomaly detection tasks can be treated as one-class classification problems in machine learning. Geometric methods can give an intuitive solution to such problems. In this paper, we propose a new geometric structure, oriented non-convex hulls, to represent decision boundaries used for one-class classification. Based on this geometric structure, a novel boundary based one-class classification algorithm is developed to solve the anomaly detection problem. Compared with traditional boundary-based approaches such as convex hulls based methods and one-class support vector machines, the proposed approach can better reflect the true geometry of target data and needs little effort for parameter tuning. The effectiveness of this approach is evaluated with artificial and real world data sets to solve the anomaly detection problem in Cyber-Physical-Production-Systems (CPPS). The evaluation results also show that the proposed approach has higher generality than the used baseline algorithms.}},

While there are different concepts in research on how to simplify the engineering process by using generic skills or capabilities of devices, run-time control is still achieved with proprietary communication protocols and commands. The concept in this paper uses skills not only in the phase of engineering but also consequently for direct and generic control of field-devices. An executable skill-metamodel therefore describes the methodological functionality which is implemented by using OPC UA due to its vendor independence as well as built-in services and information model. The implementation uses client/server-based OPC UA and the pub/sub pattern to prepare for a deterministic real-time control in conjunction with TSN, which is required by industrial automation.}},

In smart factories, network transmission has multiprotocol features of wired/wireless communication, and different data flows have different real-time requirements. In this article, a heterogeneous network architecture based on software-defined network is proposed for realizing cross-network flexible forwarding of multisource manufacturing data and optimized utilization of network resources. Subsequently, the mechanism of cross-network fusion and scheduling (CNFS) is analyzed from the perspective of high dynamic characteristics and different delay requirements of data flows. Based on this analysis, a route-aware data flow dynamic reconstruction algorithm is proposed. The proposed algorithm improves the efficiency of manufacturing data cross-network fusion, especially for multivariety and small-batch intelligent manufacturing systems. Furthermore, for meeting the bandwidth requirements of different delay flows, a delay-sensitive network bandwidth scheduling algorithm is proposed. Finally, the effectiveness of the proposed CNFS mechanism is verified using a candy packaging intelligent production line prototype platform.}},

abstract = {{The Bacillus (B.) cereus group consists of nine recognized species which are present worldwide. B. cereus play an important role in food-borne diseases by producing different toxins. Yet, only a small percentage of B. cereus strains are able to produce the heat stable cereulide, the causative agent of emetic food poisoning. To minimize the entry of emetic B. cereus into the food chain, food business operators are dependent on efficient and reliable methods enabling the differentiation between emetic and non-emetic strains.
Currently, only time-consuming cell bioassays, molecular methods and tandem mass spectrometry are available for this purpose. Thus, the aim of the present study was to establish a fast and reliable method for the differentiation between emetic/non-emetic strains by MALDI-TOF MS. Selected strains/isolates of the B. cereus group as well as other Bacillus spp. (total n = 121) were cultured on sheep blood agar for 48 h before analysis.
Subsequently, the cultures were directly analyzed by MALDI-TOF MS without prior extraction steps. The samples were measured in the mass range of m/z 800–1800 Da. Using ClinProTools 3.0 statistical software and Flex analysis software (Bruker Daltonics GmbH, Bremen, Germany), a differentiation between emetic/non-emetic isolates was possible with a rate of correct identification of 99.1% by means of the evaluation of two specific biomarkers (m/z 1171 and 1187 Da).}},

abstract = {{The fungus Stachybotrys (S.) chartarum was isolated from culinary herbs, damp building materials, and improperly stored animal forage. Two distinct chemotypes of the fungus were described that produced either high-cytotoxic macrocyclic trichothecenes (S type) or low-cytotoxic atranones (A type). Recently, two distinct gene clusters were described that were found to be necessary for the biosynthesis of either macrocyclic trichothecenes (21 SAT (Satratoxin) genes) or atranones (14 ATR (Atranone) genes). In the current study, PCR primers were designed to detect SAT and ATR genes in 19 S. chartarum chemotype S and eight S. chartarum chemotype A strains. Our analysis revealed the existence of three different genotypes: satratoxin-producing strains that harbored all SAT genes but lacked the ATR gene cluster (genotype S), non-satratoxin-producing strains that possessed the ATR genes but lacked SAT genes (genotype A), and a hitherto undescribed hybrid genotype among non-satratoxin-producing strains that harbored all ATR genes and an incomplete set of SAT genes (genotype H). In order to improve the discrimination of genotypes, a triplex PCR assay was developed and applied for the analysis of S. chartarum and S. chlorohalonata cultures. The results show that genes for macrocyclic trichothecenes and atranones are not mutually exclusive in S. chartarum. Correlation of the new genotype-based concept with mycotoxin production data shows also that macrocyclic trichothecenes are exclusively produced by S. chartarum genotype S strains.}},

  abstract     = {{Objectives
To evaluate matrix-assisted laser desorption ionisation time of flight mass spectrometry (MALDI-TOF MS) combined with the Sepsityper kit (Bruker Daltoniks GmbH, Bremen) for the direct detection of bacterial species from inoculated blood cultures from dogs and cats.
Materials and Methods
Canine and feline blood samples were inoculated with typical sepsis-causing bacteria such as Staphylococcus intermedius, Staphylococcus aureus, Streptococcus canis, Enterococcus faecalis, Escherichia coli and Pseudomonas aeruginosa at two distinct concentrations (each in triplicate), resulting in 72 blood culture bottles incubated at 37°C. Samples were comparatively analysed with MALDI-TOF MS after preparation with the Sepsityper kit and also by standard bacteriology (culturing and biochemical characterisation).
Results
Bacterial species identified from agar plates and by MALDI-TOF MS from blood culture bottles were identical for all samples. The MALDI Biotyper software (Bruker Daltoniks) correctly identified all bacterial strains from inoculated canine and feline blood with analysis indicating very good precision.
Clinical Significance
MALDI-TOF MS analysis combined with the Sepsityper kit is a reliable tool for a quick detection of veterinary-relevant bacterial species directly from blood culture bottles. This approach could reduce the time for identification of critical species to only 24 hours.}},

abstract = {{The genus Stachybotrys produces a broad diversity of secondary metabolites, including macrocyclic trichothecenes, atranones, and phenylspirodrimanes. Although the class of the phenylspirodrimanes is the major one and consists of a multitude of metabolites bearing various structural modifications, few investigations have been carried out. Thus, the presented study deals with the quantitative determination of several secondary metabolites produced by distinct Stachybotrys species for comparison of their metabolite profiles. For that purpose, 15 of the primarily produced secondary metabolites were isolated from fungal cultures and structurally characterized in order to be used as analytical standards for the development of an LC-MS/MS multimethod. The developed method was applied to the analysis of micro-scale extracts from 5 different Stachybotrys strains, which were cultured on different media. In that process, spontaneous dialdehyde/lactone isomerization was observed for some of the isolated secondary metabolites, and novel stachybotrychromenes were quantitatively investigated for the first time. The metabolite profiles of Stachybotrys species are considerably influenced by time of growth and substrate availability, as well as the individual biosynthetic potential of the respective species. Regarding the reported adverse effects associated with Stachybotrys growth in building environments, combinatory effects of the investigated secondary metabolites should be addressed and the role of the phenylspirodrimanes re-evaluated in future research.}},
