abstract = {{The Paper presents the concept of an experimental plant, which is to be built within the framework of the research project DC-INDUSTRIE2. This system consists of a multitude of electrical drives, various uni- and bidirectional rectifiers, several decentralized energy suppliers and several energy storage units. All devices in this system are coupled via a common DC Grid. DC Grids allow a very simple bidirectional energy exchange between the connected devices. Therefore, braking energy in electric drives, for example, can be used without additional effort and the integration of decentralized energy supplier systems such as photovoltaic (PV) or combined heat and power plants (CHP) as well as energy storage systems is much easier than in AC Grids. The purpose of the experimental plant is to electrically emulate a typical production machine or process and to investigate the real occurring currents and energy flows in the DC Grid. A total of 11 drive sets are available for this purpose, each consisting of a test machine and a load machine. The modeling of the actual production machine is done in MATLAB/Simulink. Speed and torque curves are calculated here and transmitted to the drive inverters via fieldbus. Thus, not only real mechanical quantities are obtained at the shaft, but also the currents in the DC Grid correspond to those of a real plant. Therefore it can be investigated, for example, how a production machine converted from AC to DC power would behave without having to build it. Furthermore, the effects of storage connections and grid disturbances can be realistically investigated.}},

abstract = {{Material flows and energy flows can be correlated with sufficient data, e.g. on production energies, annual production quantities and degrees of dissipation with temperature increases in the atmosphere, volumes of molten ice or sea level increases, as well as with probability statements, information densities and management recommendations. All these quantities can be described by the comprehensive term entropy.
In order to consider the efficiency of material and energy flows, the difficulty to understand the concept of entropy with its different definitions can be summarized simply and easily in a model related to ice cubes.
The quality of a model containing percentage probability statements, statements on dissipation in material flow models in connection with statements on information density and its description by the ice cube model is still to be determined in practice in suitable material flow models. Such projects should show the types of mathematical correlations between dissipation degrees, entropy increase, increase of molten ice and sea level rise.

abstract = {{Digitalization crumbled the infrastructure of organizational journalism. Old-line institutions took radical measures to survive. A new age began... For some; this was a disaster for journalism; for others, it was the commencement of a golden age. It was always up to public. Public is the determinantfactor for quality journalism. Being more conscious with technological means; participation in journalistic activities and demand on quality might lead to a ‘’better journalism’’. Some emergant business models support this approach; one of them being slow journalism. Journalism; slowly and consciously, but how does it work? Can slow journalism help people navigate in the post-truth era or is it just a product of a life style politics? In order to find that; a qualitative media content analysis is performed between two journalistic products. The results show that; the selected slow journalism product offer not a ‘’richer’’ content. But slow journalism has many forrms and it is not yet clearly defined.Journalism is ideally a noble proffesion. The noble ideas and purposes of journalism are used very often for marketing purposes. With qualitative media content analysis, these noble purposes can be observed, evaluated and new strategies can be developped to improve them.}},

abstract = {{In 2019 DOCOMOMO Germany and the Detmold School of Architecture and Interior Architecture at Ostwestfalen-Lippe, University of Applied Sciences (TH OWL)

with the Erasmus+-Project ‘Reuse of Modernist Buildings‘ (RMB) organized the 16th DOCOMOMO Germany and 3rd RMB Conference. The international conference

in Berlin took place on the occasion of the 100th anniversary of the Bauhaus and as an opportunity to discuss the significance of modernity in the 21st century.

The conference focus lies on the concepts, visions, and impulses emanating from Modern Movement and how they can be related to today’s social, economic, cultural

and in particular creative issues.

This second Docomomo publication includes a selection of eleven papers that were not included in the original online conference proceedings (http://www.rmb-eu.

com/publications/). The papers witness in a particular way the dominating themes and typologies of Modern Movement. They also demonstrate manifold reuse and

conservation approaches—conceptually, aesthetically and technically. They are expression of the intensive investigation and documentation efforts of members and

supporters of DOCOMOMO together with the academic and professional community. }},

abstract = {{Obsolescence and urban decay are usual attributes of the Modern Movement buildings and areas worldwide, especially of the post-war large-scale housing settlements. Therefore, the question of reuse and improvement guidelines for these settlements is becoming increasingly important. The paper addresses this issue, taking New Belgrade housing blocks as a case study. The current condition of the New Belgrade blocks that, indeed, can be characterised as obsolete and not adequately maintained, even degraded, is nevertheless a perfect platform for reading of the architecture and the space values, and how it was changing during the time. The study aim was reading the unforeseen impulses of modernism: identification and mapping of socio-spatial relations in environment (reactions on and interventions in space that were generated during the time), reading and interpreting them as impulses of the user behaviours and lifestyles, and further proposing future transformation tactics using the mapped elements and principles. The research framework was a workshop-seminar organized by the authors at the Faculty of Architecture in Belgrade. Using this research tactic, the particular elements and spaces within the blocks were mapped, identified, classified and systematized according to their flexibility to react and accept contemporary impulses of life. The study reveals common spaces of the New Belgrade blocks, or spaces between private and public, as the key elements in managing the urban decay and prompting adaptation, and hence investigates on the potential of their adaptive reuse initiating transformation of the whole area, and further achieving its attractiveness, openness and better accessibility. }},

abstract = {{This bachelor thesis explores the adaptive reuse of the former Telekom high-rise at Kesselbrink in Bielefeld, a vacant modernist office tower awaiting new purpose. The project proposes a mixed-use transformation that reinterprets the building as a “city within the city,” inspired by Le Corbusier’s Unité d’Habitation in Marseille. The existing structure is strategically reduced to its essential load-bearing concrete frame and floor slabs, preserving the structural skeleton while enabling radical spatial reconfiguration.
Prefabricated timber modules are inserted and cantilevered between the reinforced-concrete columns, extending the usable floor area and introducing a warm, contemporary material contrast to the original façade. This intervention not only increases density and flexibility but also redefines the tower’s appearance, giving it a cosmopolitan and forward-looking architectural identity. The ground floor is conceived as an open and publicly accessible urban platform, activating the surrounding plaza and strengthening social interaction. Upper floors are redesigned as affordable and attractive living spaces for young residents, encouraging a diverse and vibrant community within the building.
A newly designed park-like rooftop landscape provides recreational space and panoramic views over the city, enhancing environmental quality and urban connectivity. By combining structural preservation, modular timber construction, public programming, and residential reuse, the proposal demonstrates how high-rise retrofitting can generate social, ecological, and economic value simultaneously. The project positions adaptive reuse as a viable strategy for sustainable urban regeneration and architectural longevity.}},

This study sought to replicate and extend this line of research by investigating whether and under what conditions preschool children generalise EC effects from conditioned to novel stimuli. Specifically, two experiments were conducted in which 3- to 6-year-old children (N = 139) observed the pairing of two cartoon characters with two positive and negative images. Afterwards, children evaluated the paired characters as well as other novel characters, which were not previously presented, that varied systematically in their perceptual similarity to the conditioned stimuli (Experiments 1 & 2), or that could be grouped by a categorisation rule (Experiment 2). It was found that children generalised attitudes to perceptually similar stimuli, but not to stimuli related by a categorisation rule, despite evidence for rule-learning. Implications of these findings for attitude development are discussed.}},

Therefore, in the present study we employed a single-trial, intentional learning procedure that paired neutral with valenced words while recording ERP activity, and measured subsequent memory and subsequent attitudes for the pre-experimentally neutral words immediately following learning. Using traditional as well as single-trial ERP analyses, we found that frontal slow wave (FSW) activity, elicited while stimuli were being paired, was associated with both subsequent memory for the word pairs and subsequent attitudes towards the pre-experimentally neutral words. Specifically, FSW activity during the pairing of neutral with positive words was related to more positive subsequent attitudes, while during the pairing of neutral with negative words, it was associated with more negative subsequent attitudes, towards the pre-experimentally neutral words. Given that this FSW activity was also related to successful subsequent episodic memory retrieval for the word pairs, these findings provide evidence that the transfer of valence may depend on a process that supports associative episodic encoding during word paring. Further, a single-trial, mixed-effects model indicated that the relationship between encoding FSW activity and subsequent attitudes depended on the strength of the episodic memory trace. Thus, the present study provides novel evidence that a common mechanism contributes to both episodic memory encoding and affective attitude formation.}},

Participants experienced personal RD as the difference between own rewards and the rewards of a fictitious other player. Going beyond previous research, three yet-unexplored moderators of the RD-aggression link were experimentally tested: In Experiment 1 (N = 157), we tested the effect of the scarcity of resources one is deprived of, and the intensity of the RD experience in terms of the magnitude of the disadvantaged comparison. In Experiment 2 (N = 195), we investigated whether aggressive behavior is influenced by imposing possible or actual sanctions (i.e., costs) for aggression. The results show effects on aggressive behavior toward the other player that were mediated by the personal perception of RD and that only actual but not possible costs eliminated this indirect effect. Implications for the aggression-related behavioral consequences of RD are discussed.}},

Here, we examined whether and how the stress-induced release of cortisol, following the socially-evaluated cold pressor test, influenced the acquisition of preferences in an evaluative conditioning (EC) procedure. We found that when the stressor preceded the evaluation phase, cortisol responders showed decreased evaluative conditioning effects. By contrast, impairing effects of a stressor-induced cortisol release before encoding were not found. Moreover, explicit memory was not found to be affected by the stressor or its timing. Implications of the timing-dependent effects of stress-induced cortisol release on EC and the relation between stress and associative memory are discussed.}},

The success of the production system stems from its methods and its focus on human factors. For some years, production research has focused on the topic of digital manufacturing. This technology-oriented approach is pursued quite independently of the Toyota Production System. As a result, technical solutions may prove incompatible with the Lean philosophy. Therefore, operational practice must link the Lean philosophy with new technologies in order to make work processes and material flows productive and ergonomic simultaneously. As a part of their education in industrial engineering at the Ostwestfalen-Lippe University of Applied Sciences and Arts, students learn all current Lean methods by means of business games. One of these has been supplemented with information technology components. The objective of this article is to introduce this business game and to explain its didactic concept.}},

With the help of assistance systems, the process of information absorption and processing can be designed in a more economical and human-oriented way. Despite the potentials of such systems, companies pay too little attention to the informational design of assembly systems, resulting in a number of significant deficits in information management. To identify such deficits in operational practice, a questionnaire has been developed. This allows the potential use of informational assistance systems for an assembly system to be estimated. In this article, results from an exploratory factor analysis of the developed instrument will be presented and interpreted as a basis for the further development of the questionnaire.}},

abstract = {{Flocculation filtration in deep bed is one of the techniques to achieve advanced phosphorus removal. In addition to particle retention and phosphorus removal, granular activated carbon may serve as a filtration medium but also as an adsorption bed for micropollutants. The aim of the study was to investigate the simultaneous elimination of total phosphorus (TP) and selected micropollutants. In the first part of the study the target phosphorus threshold of 0.2 mg/L TP in the filtrate was achieved in a dual-bed media filter with anthracite and sand up to filter velocities of 16 m/h. By using a detailed sampling protocol and headloss curves, the influence of different operation modes on retention could be identified. Afterwards, the filter media were replaced by three types of GAC. In all filter beds substantial differences were found in the mode and efficiency of particle retention. This led to different filter run times, which is one of the main parameters for efficient and economical filter operation. Specific curves were generated for the different bed materials showing the dependency of filter run time on the specific load of suspended solids. Elimination of micropollutants in a GAC 4 × 8 mesh filter bed was low due to coarse grains and low empty bed contact time (EBCT). The application of GAC 8 × 14 mesh filter bed resulted in high elimination of most micropollutants. These results can be applied when choosing filter bed material. This may involve a compromise between filter run time and adsorption of micropollutants.}},

  abstract     = {{Increasing heterogeneity of industrial network systems is a fact and the chances that in the future one communication standard will be able to fulfill the requirements of all possible applications are utopian. With the increasing number of communication systems, their management, configuration, and maintenance become a significant issue. Additionally, due to the increasing amount of network services and traffic, the management of available network resources and the possibility of delivering certain levels of communication quality of service, especially across different network solutions becomes a big challenge. Therefore, in this paper a Controller of Controllers (CoC) concept for management of heterogeneous industrial networks is proposed. The concept is designed to support still widely spread legacy fieldbus systems, different Ethernet-based industrial solutions, and potentially upcoming network technologies. The goal is achieved by leveraging state-of-the-art architectural concepts such as software-defined networks, which allows for integration of abstract models in network management.   abstract     = {{
Since the 1960s, atopic dermatitis has seen a steady increase in prevalence in developed countries. Most often, the onset begins at an early age and many patients are very young children. Due to their young age, their parents are forced to take over handling of the disease. As a consequence, atopic dermatitis places a high burden not only on affected children, but also on their parents and siblings, limiting human flourishing of a whole family. Therefore, the described research area calls for a possibility-driven approach that looks beyond mere problem-solving while building on existing support possibilities and creating new ones. This paper presents atopi as a result of such a possibility-driven approach. It incorporates existing patient education and severity scoring into an extensive service, adding new elements to turn necessary practices into joyful experiences, to create feelings of relatedness and to increase perceived self-efficacy, thus is suitable to enable human flourishing.
}},

  abstract     = {{
A non-negligible proportion of seniors rarely uses new technology. Far too often, these rather tech unexperienced seniors struggle with new soft- or hard-ware, that do not provide easy access. As a result, some seniors avoid or completely desist to further use “these modern technologies” and consequently miss out on the benefits of digital age. Game-inspired design is supposed to be a promising way to overcome some perceived barriers of seniors by providing hedonic value during early interaction. Previous research has shown that game-inspired design is suitable to motivate seniors’ use of Health-IT. To investigate its potential to facilitate the use of information and communication technology (ICT), an experimental study was conducted. The study investigated the appeal of various gamification features, which were embedded in a prototype of an ICT learning software and compared it to a non-gamified version of the software. Results indicate that the concept of gamified ICT learning software appeals to seniors in general, but that the acceptance of different gamification features is quite diverse. A clear-cut superiority of adding gamification to the software was not found. After interacting with both software versions, seniors in around equal parts preferred either the non-gamified version, the gamified version, or could not decide. Those seniors that clearly favored the gamified version were particularly fond of continuous positive feedback and receiving rewards for each task they have accomplished. Whereas the remaining seniors rather disliked the intensive endorsement through these two features and decided against the gamified version, albeit they liked many of the other game features. Our results underline the necessity of following a user-centered design approach when developing game-inspired applications, and the need for an individualized use of gamification elements to meet the needs of the heterogeneous group of senior users.
}},

In the early 1990s the Aleppo municipality initiated the project for the Rehabilitation of the Old City with the German Technical Cooperation Agency (GTZ) as a partner. As part of the process, a Development Plan was issued as a general framework including ten strategies for upgrading the historical urban fabric in line with the sustainable development. Despite implementation of these strategies, the physical fabric of the Old City has continued to deteriorate, and the demographic transition has continued to increase until the escalation of the Syrian Civil War in 2011. The paper analyses the development plan{\textquoteright}s strategies in the Old City of Aleppo, mainly those related to housing aspects. It evaluates the effectiveness of these strategies based on the concept of good urban governance that intersects with the Sustainable Development Goals (SDGs) aiming to overcome the administrative gaps to rebuild the Old City more sustainably after the Syrian Civil War.}},

As brand representatives, service employees play a critical role in these recollections. Their behaviour might inform job seekers’ understanding of the employer brand, thereby affecting their application decision. The impact of the service employee’s behaviour might be even more impactful if this behaviour is branded, meaning that the employee’s appearance and manner are representative of the brand values. In this paper, we analyse whether and how branded service employee behaviour affects job seekers’ application intentions. We argue that branded service employee behaviour provides two opposing signals: it contributes to consistent evaluations of the brand, leading to more brand trustworthiness and enhancing job seekers’ application intentions. However, it may also be perceived as inauthentic, reducing job seekers’ application intentions. Findings from a scenario experiment provide evidence of both effects.}},

For a gentle dimensioning of a pasteurization plant the knowledge of the kinetic figures is important. Activation energy, reaction order and pre-exponential factor of the L-ascorbic acid degradation in a model solution, apple, orange and black currant juice were determined. Lines of equal effects, which indicate different time-temperature combinations for the degradation, could be derived and compared with the lethal effect on microorganisms. The activation energies were located in the area of 25 to 44 kJ/mol for all samples except of orange juice (74 kJ/mol) in the range of 40-90 ?C with a zeroth reaction order. Based on these values, the lines of equal effects showed a lesser degradation at higher temperatures and shorter holding times even in the typical setting range of pasteurization plants.}},

abstract = {{Continuous mashing provides advantages compared to conventional batch-wise mashing in terms of space time yield. The majority of fermentable sugars are generated during the so-called “β-amylase rest” (62–64 ◦C). These low molecular sugars are fermented later in the brewing process by yeasts and therefore determine the beer attenuation degree. Biological malt variations complicate the application of a continuous system in industrial scale particularly concerning targeted quality parameters. The aim is the prediction of sugar formation from process parameters for a real time control system. Therefore, a semi-empirical model for sugar formation in a continuous stirred tank reactor (CSTR) system was developed under incorporation of the residence time distri- bution (RTD). The here presented model, which focuses on the “β-amylase rest”, is able to predict fermentable sugar concentrations in the continuous “β-amylase rest” with sufficient accuracy, in contrast to models that only use the flow rate and the reactor volume to determine the reaction time. However, the precision and trueness depend on the quality of the empirical data acquired previously in laboratory experiments for the selected temperature and raw material quality.}},

abstract = {{Near infrared spectroscopy in combination with a transflection probe was investigated as inline measurement in a continuous flash pasteurizer system with a sugar-water model solution. Robustness and reproducibility of fluctuations of recorded spectra as well as trueness of the chemometric analysis were compared under different process parameter settings. Variable parameters were the flow rate (from laminar flow at 30 L/h to turbulent flow at 90 L/h), temperature (20 to 100 degrees C) and the path length of the transflection probe (2 and 4 mm) while the pressure was kept constant at 2.5 bar. Temperature and path length were identified as the most affecting parameters, in case of homogenous test medium. In case of particle containing systems, the flow rate could have an impact as well. However, the application of a PLS model, which includes a broad temperature range, and the correction of prediction results by applying a polynomial regression function for prediction errors, was able to compensate these effects. Also, a path length of 2 mm leads to a higher accuracy. The applied strategy shows that by the identification of relevant process parameters and settings as well as the establishment of a compensation strategy, near infrared spectroscopy is a powerful process analytical tool for continuous flash pasteurization systems.}},

abstract = {{In temperate lakes, it is generally assumed that light rather than temperature constrains phytoplankton growth in winter. Rapid winter warming and increasing observations of winter blooms warrant more investigation of these controls. We investigated the mechanisms regulating a massive winter diatom bloom in a temperate lake. High frequency data and process-based lake modeling demonstrated that phytoplankton growth in winter was dually controlled by light and temperature, rather than by light alone. Water temperature played a further indirect role in initiating the bloom through ice-thaw, which increased light exposure. The bloom was ultimately terminated by silicon limitation and sedimentation. These mechanisms differ from those typically responsible for spring diatom blooms and contributed to the high peak biomass. Our findings show that phytoplankton growth in winter is more sensitive to temperature, and consequently to climate change, than previously assumed. This has implications for nutrient cycling and seasonal succession of lake phytoplankton communities. The present study exemplifies the strength in integrating data analysis with different temporal resolutions and lake modeling. The new lake ecological model serves as an effective tool in analyzing and predicting winter phytoplankton dynamics for temperate lakes.}},

We applied a two-dimensional hydrodynamic model to explore the response of the thermal structure in Germany's largest drinking water reservoir, Rappbode Reservoir, to future climate projections and different water withdrawal strategies. We used projections for representative concentration pathways (RCP) 2.6, 6.0 and 8.5 from an ensemble of 4 different global climate models. Simulation results showed that epilimnetic water temperatures in the reservoir strongly increased under all three climate scenarios. Hypolimnetic temperatures remained rather constant under RCP 2.6 and RCP 6.0 but increased markedly under RCP 8.5. Under the intense warming in RCP 8.5, hypolimnion temperatures were projected to rise from 5 °C to 8 °C by the end of the century. Stratification in the reservoir was projected to be more stable under RCP 6.0 and RCP 8.5, but did not show significant changes under RCP 2.6. Similar results were found with respect to the light intensity within the mixed-layer. Moreover, the results suggested that surface withdrawal can be an effective adaptation strategy under strong climate warming (RCP 8.5) to reduce surface warming and avoid hypolimnetic warming. This study documents how global scale climate projections can be translated into site-specific climate impacts to derive adaptation strategies for reservoir operation. Moreover, our results illustrate that the most intense warming scenario, i.e. RCP 8.5, demands far-reaching climate adaptation while the mitigation scenario (RCP 2.6) does not require adaptation of reservoir management before 2100.}},

abstract = {{Metalimnetic oxygen minima are observed in many lakes and reservoirs, but the mechanisms behind this phenomena are not well understood. Thus, we simulated the metalimnetic oxygen minimum (MOM) in the Rappbode Reservoir with a well-established two-dimensional water quality model (CE-QUAL-W2) to systematically quantify the chain of events leading to its formation. We used high-resolution measured data to calibrate the model, which accurately reproduced the physical (e.g. water level and water temperature), biogeochemical (e.g. nutrient and oxygen dynamics) and ecological (e.g. algal community dynamics) features of the reservoir, particularly the spatial and temporal extent of the MOM. The results indicated that around 60% of the total oxygen consumption rate in the MOM layer originated from benthic processes whereas the remainder originated from pelagic processes. The occurrence of the cyanobacterium Planktothrix rubescens in the metalimnion delayed and slightly weakened the MOM through photosynthesis, although its decaying biomass ultimately induced the MOM. Our research also confirmed the decisive role of water temperature in the formation of the MOM since the water temperatures, and thus benthic and pelagic oxygen consumption rates, were higher in the metalimnion than in the hypolimnion. Our model is not only providing novel conclusions about the drivers of MOM development and their quantitative contributions, it is also a new tool for understanding and predicting ecological and biogeochemical water quality dynamics.}},

abstract = {{Vapor phase hydrogen peroxide (H2O2) can be utilized to inactivate murine norovirus (MNV), a surrogate of human norovirus, on surface areas. However, vapor phase H2O2 inactivation of virus on fruits and vegetables has not been characterized. In this study, MNV was used to determine whether vaporized H2O2 inactivates virus on surfaces of various fruits and vegetables (apples, blueberries, cucumbers, and strawberries). The effect of vapor phase H2O2 decontamination was investigated with two application systems. Plaque assays were performed after virus recovery from untreated and treated fresh produce to compare the quantity of infective MNV. The Mann-Whitney U test was applied to the test results to evaluate the virus titer reductions of treated food samples, with significance set at P <= 0.05. The infective MNV populations were significantly reduced on smooth surfaces by 4.3 log PFU (apples, P < 0.00001) and 4 log PFU or below the detection limit (blueberries, P = 0.0074) by treatment with vapor phase H2O2 (60 min, maximum of 214 ppm of H2O2). Similar treatments of artificially contaminated cucumbers resulted in a virus titer reduction of 1.9 log PFU. Treatment of inoculated strawberries resulted in 0.1and 2.8-log reductions of MNV. However, MNV reduction rates on cucumbers (P = 0.3809) and strawberries (P = 0,7414) were not significant. Triangle tests and color measurements of untreated and treated apples, cucumbers, blueberries, and strawberries revealed no differences in color and consistency after H2O2 treatment. No increase of the H2O2 concentration in treated fruits and vegetables compared with untreated produce was observed. This study reveals for the first time the conditions under which vapor phase H2O2 inactivates MNV on selected fresh fruit and vegetable surfaces.}},

abstract = {{Modern product development utilizes both Product Lifecycle Management (PLM) and Application Lifecycle Management (ALM). PLM addresses the hardware lifecycle of a product, whereas ALM addresses the software lifecycle. In recent years, industry and academia have developed several PLM/ALM integration concepts to realize efficient management of the product lifecycle across all domains. However, the solutions available in practice are typically vendor-driven. Therefore, they are not generally applicable even if standards such as OSLC (Open Services for Lifecycle Cooperation) are applied.

The consortium "Plattform Industrie 4.0" has recently introduced a standardized digital representation of an asset (e.g., a smart product): the Asset Administration Shell (AAS). The AAS has the potential to integrate PLM/ALM data sets in a single product model and hence to provide a generally applicable interface for PLM/ALM integration. However, until now there has not been a concept to prove this potential. The aim of this work is to develop such new strategies (named Plm4AAS) using AAS submodels. This article explains the semi-automatic generation of PLM/ALM submodels and how to link elements between these submodels. The AASX Package Explorer, an AAS management software tool, is used to demonstrate the results. The article finishes with a discussion about the potential of the AAS as a standardized concept for PLM/ALM integration. (C) 2020 The Authors. Published by Elsevier B.V. Although a multitude of solutions has been applied to this modern Industry 4.0 problem, it remains an open research issue to identify the key characteristics subjacent to an anomaly, sc. generate hypothesis as to why they appear. In recent years, machine learning models have been regarded as universal solution for a wide range of problems. While most of them suffer from non-self-explanatory representations, Gaussian Processes (GPs) deliver interpretable and robust statistical data models, which are able to cope with unreliable, noisy, or partially missing data. Thus, we regard them as a suitable solution for detecting and appropriately representing anomalies and their respective characteristics. In this position paper, we discuss the problem of automatic and interpretable anomaly detection by means of GPs. That is, we elaborate on why GPs are well suited for anomaly detection and what the current challenges are when applying these probabilistic models to large-scale production data.}},

Authors propose a novel approach using the hot-melt extrusion technique to prepare delayed release dosage forms via a continuous manufacturing process, a new trend in the pharmaceutical industry. A full factorial design was employed to correlate input variables, including stearic acid (SA) content, drug content, and pellet size with drug release properties of the pellets. PLS fit method suitably elaborated the relationship between input and output variables with reasonably good fit and goodness of prediction. All three input factors influenced drug release in enzyme-free simulated gastric fluid (SGF) after 120 min; however, SA content did not significantly affect drug dissolution in the enzyme-free simulated intestinal fluid (SIF). An optimized formulation and design space were determined by overlaying multiple contours established from regression equations. The continuous manufacturing process was successfully monitored using inline near-infrared (NIR) and inline particle size analysis, with drug load and pellet size being well-controlled within the design space. The obtained pellets released less than 5% after 120 min in SGF and more than 85% and 95% after 30 min and 45 min, respectively, after switching to SIF. (C) 2020 American Pharmacists Association (R).

abstract = {{Stachybotrys (S.) chartarum had been linked to severe health problems in humans and animals, which occur after exposure to the toxic secondary metabolites of this mold. S. chartarum had been isolated from different environmental sources, ranging from culinary herbs and improperly stored fodder to damp building materials. To access the pathogenic potential of isolates, it is essential to analyze them under defined conditions that allow for the production of their toxic metabolites. All Stachybotrys species are assumed to produce the immunosuppressive phenylspirodrimanes, but the highly cytotoxic macrocyclic trichothecenes are exclusively generated by the genotype S of S. chartarum. In this study, we have analyzed four genotype S strains initially isolated from three different habitats. We grew them on five commonly used media (malt-extract-agar, glucose-yeast-peptone-agar, potato-dextrose-agar, cellulose-agar, Sabouraud-dextrose-agar) to identify conditions that promote mycotoxin production. Using LC-MS/MS, we have quantified stachybotrylactam and all S-type specific macrocyclic trichothecenes (satratoxin G, H, F, roridin E, L-2, verrucarin J). All five media supported a comparable fungal growth and sporulation at 25 °C in the dark. The highest concentrations of macrocyclic trichothecenes were detected on potato-dextrose-agar or cellulose-agar. Malt-extract-agar let to an intermediate and glucose-yeast-peptone-agar and Sabouraud-dextrose-agar to a poor mycotoxin production. These data demonstrate that the mycotoxin production clearly depends on the composition of the respective medium. Our findings provide a starting point for further studies in order to identify individual components that either support or repress the production of mycotoxins in S. chartarum.}},

  abstract     = {{Background
Borrelia burgdorferi is a tick-borne spirochete that causes Lyme borreliosis (LB). After an initial tick bite, it spreads from the deposition site in the dermis to distant tissues of the host. It is generally believed that this spirochete disseminates via the hematogenous route. Borrelia persica causes relapsing fever and is able to replicate in the blood stream. Currently the exact dissemination pathway of LB pathogens in the host is not known and controversially discussed.
Methods
In this study, we established a strict intravenous infection murine model using host-adapted spirochetes. Survival capacity and infectivity of host-adapted B. burgdorferi sensu stricto (Bbss) were compared to those of B. persica (Bp) after either intradermal (ID) injection into the dorsal skin of immunocompetent mice or strict intravenous (IV) inoculation via the jugular vein. By in vitro culture and PCR, viable spirochetes and their DNA load in peripheral blood were periodically monitored during a 49/50-day course post-injection, as well as in various tissue samples collected at day 49/50. Specific antibodies in individual plasma/serum samples were detected with serological methods.
Results
Regardless of ID or IV injection, DNA of Bp was present in blood samples up to day 24 post-challenge, while no Bbss was detectable in the blood circulation during the complete observation period. In contrast to the brain tropism of Bp, Bbss spirochetes were found in ear, skin, joint, bladder, and heart tissue samples of only ID-inoculated mice. All tested tissues collected from IV-challenged mice were negative for traces of Bbss. ELISA testing of serum samples showed that Bp induced gradually increasing antibody levels after ID or IV inoculation, while Bbss did so only after ID injection but not after IV inoculation.
Conclusions
This study allows us to draw the following conclusions: (i) Bp survives in the blood and disseminates to the host’s brain via the hematogenous route; and (ii) Bbss, in contrast, is cleared rapidly from the blood stream and is a tissue-bound spirochete.}},

abstract = {{The genus Borrelia comprises vector-borne bacterial pathogens that can severely affect human and animal health. Members of the Borrelia burgdorferi sensu lato species complex can cause Lyme borreliosis, one of the most common vector-borne diseases in the Northern hemisphere. Besides, members of the relapsing fever group of spirochetes can cause tick-borne relapsing fever in humans and various febrile illnesses in animals in tropical, subtropical and temperate regions. Borrelia spp. organisms are fastidious to cultivate and to maintain in vitro, and therefore, difficult to work with in the laboratory. Currently, borrelia identification is mainly performed using PCR and DNA sequencing methods, which can be complicated/frustrating on complex DNA templates and may still be relatively expensive. Alternative techniques such as matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) are not well established for Borrelia spp., although this technique is currently one of the most used techniques for rapid identification of bacteria in microbiological diagnostic laboratories. This is mainly due to unsatisfactory results obtained by use of simple sample preparation techniques and medium-contamination obscuring the mass spectra. In addition, comprehensive libraries for Borrelia spp. MALDI-TOF MS have yet to be established. In this study, we developed a new filter-based chemical extraction technique that allows measurement of high quality Borrelia spp. spectra from less than 100,000 bacteria per spot in MALDI-TOF MS. We used 49 isolates of 13 different species to produce the largest mass-library for Borrelia spp. so far and to validate the protocol. The library was successfully established and identifies >96% of used isolates correctly to species level. Cluster analysis on the sum spectra was applied to all the different isolates, which resulted in tight cluster generation for most species. Comparative analysis of the generated cluster to a phylogeny based on concatenated multi-locus sequence typing genes provided a surprising homology. Our data demonstrate that the technique described here can be used for fast and reliable species and strain typing within the borrelia complex.}},

abstract = {{Antibodies represent an important element in the adaptive immune response and a major tool to eliminate microbial pathogens. For many bacterial and viral infections, efficient vaccines exist, but not for fungal pathogens. For a long time, antibodies have been assumed to be of minor importance for a successful clearance of fungal infections; however this perception has been challenged by a large number of studies over the last three decades. In this review, we focus on the potential therapeutic and prophylactic use of monoclonal antibodies. Since systemic mycoses normally occur in severely immunocompromised patients, a passive immunization using monoclonal antibodies is a promising approach to directly attack the fungal pathogen and/or to activate and strengthen the residual antifungal immune response in these patients.}},
